Tablet formulation by designed experiments

Find the best binder and disintegrant levels for a tablet using a designed experiment and a response surface.

The brief

Formulators often change one ingredient at a time and miss how ingredients interact. Plan a designed experiment (factorial or central composite) for binder and disintegrant levels, fit a model for hardness, friability and disintegration time, and find the region where every quality target is met. Good looks like: a proper design table, model checks, a contour plot of the design space and a recommended formula with a margin of safety.

What you will build

  1. Choose factors, levels and quality targets
  2. Build the experimental design
  3. Collect or simulate the responses
  4. Fit regression models and check them
  5. Plot contours and find the design space
  6. Recommend and justify a formula
